This kit was based on Capture-ELISA detection method. Anti Guinea pig IgE was pre-coated onto the 96-well plate. The biotin conjugated antigen was used as the detection antigen. Then, biotin detection antigen was added to bind with OVA sIgE conjugated on Anti Guinea pig IgE. Excess conjugate and unbound sample or standard are washed from the plate, and HRP-Streptavidin (SABC) is added to each microplate well and incubated. Then TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm. The concentration of OVA sIgE in the samples is then determined by comparing the OD of the samples to the standard curve. The concentration of the target substance was inversely proportional to the OD450 value.
