Background

Staphylococcus aureus V8 protease (Endoproteinase Glu-C) belongs to the serine protease family and can specifically hydrolyze carboxy-terminal peptide bonds of glutamic acid (Glu) or aspartate (Asp) residues. Its specificity is directly affected by the composition of the buffer, recognizing and cutting Glu carboxy-terminal peptide bonds in NH4HCO3 of pH7.8 and CH3COONH4 of pH4.0, recognizing and cutting Glu or Asp carboxy-terminal peptide bonds in phosphate buffers of pH7.8,but the hydrolysis rate of Glu was higher than that of Asp. Glu-C has been reported to be active in 0.2% SDS (sodium lauryl sulfate) and 4.0M urea.

Components

Supplied in dry format from Tris-HCl and sodium chloride buffer

Protocol

For the digestion of peptides or proteins, the ratio of enzyme to substrate between is 1:20 and 1:100 (w/w) for recommended. Dissolve the peptide or protein and was enzymolysis in 100 mM NH4HCO3, pH 7.8 or 100 mM Tris-HCl, pH 7.8, the recommended incubation time is 2-18 hours at 37°C, depending on the temperature and the ratio of enzyme to substrate.

Guidelines

Repeated freeze/thawing is not recommended.

Unit Definition

Using 2-Ph-leu-Glu-4-Na as substrate, the amount of enzyme required to produce 1μmol 4-Nitroaniline in 1 minute at pH7.8, 25℃ was 1unit.

Great book!
Format: Paperback
This book is a great complement to my book "The Secret To Getting A Job After College". While I cover the details about resumes and cover letters, this book does a very effective job addressing issues related to determining what you should think about doing after college and the author also has a valuable section on how to prepare your elevator speech. I recommend it for everyone who is in college or has graduated in the past few years and I now consider it required reading for many of my students.

Career Coach Recommended!
Format: Paperback
As a Career Coach, I find this book to be a great resource for college students and recent grads who are interested in preparing for a rewarding career after graduation. Lindsey Pollack is both knowledgeable and encouraging, and presents multiple strategies for students to be career-ready in today's competitive job market. A few topics include interview preparation, networking, service events, and blogging. A great read for the millennial student or job seeker, regardless of whether they have identified their career path or not.

This is by far the most helpful book I have actually enjoyed reading for school
Format: Paperback
I had to purchase this book for my pre-professional seminar class. This is by far the most helpful book I have actually enjoyed reading for school. This book covers a wide range of topics and is very specific on the steps take to to reach your career goals. As a student who is undeclared I found this book very valuable. I also like the style of writing, the author talks to you like a friend.

Now what I was looking for
Format: Paperback
I was looking more for advice on getting from a major with no clear career applications(ie. not doctor, lawyer or teacher) to a career that I would be able to live with and perhaps enjoy. This guide helps you more if you have a pretty clear idea what you want to do and are ready to make a full time job of finding your career, especially if you are not willing to settle for "paying the bills" or "get out of the parents' house". For those of you lucky enough to have not picked a major yet, just remember that picking a major needs to corrolate to getting a job, not just "learning stuff". For everyone else, if you have a dream, seems like this book will help you get there. If you don't yet have a dream, I would strongly suggest tackling that area first. This book mainly just suggests getting a career profile(which is more of other people telling you what you should do, instead of you finding out what you want to do). There's a lot of really helpful and very current information in here. You have to be ready to jump on it though.

Outstanding! An absolute must for college students
Format: Paperback
I picked this book up hoping it would prepare me for the job market, which I was already pretty confident about. It was incredibly eye opening, and enlightened me about dozens of things I was overlooking--like using Facebook as my "professional friend" and getting a linkedin account. It was a little stressful seeing how much I could do, but I've been working through the tips one at a time and its really built momentum in my professional brand even as an undergraduate. Now, I have something to do to build my career everyday.
