VDAC1, VDAC2, and VDAC3 are three closely related ~30 kDa β-barrel porins embedded in the mitochondrial outer membrane where they form voltage-dependent anion channels that collectively serve as the primary gatekeepers for metabolite flux—allowing ATP/ADP, NADH, ion, and other small-molecule exchange between cytosol and mitochondria—while also acting as hubs for Ca²⁺ signaling, apoptotic protein binding, and interaction with hexokinase, creatine kinase, Bcl-2 family proteins, and cytoskeletal elements; despite high sequence similarity, isoforms show tissue-specific expression (VDAC1 ubiquitous, VDAC2 enriched in heart/testis and uniquely anti-apoptotic via Bax inhibition, VDAC3 prominent in testis and cilia with redox-sensitive cysteines linked to sperm motility and ROS regulation), distinct electrophysiological gating properties, and differential roles in oxidative stress, mitochondrial dynamics, and apoptosis, making the trio essential for bioenergetics, cell survival decisions, and increasingly recognized as therapeutic targets in cancer, neurodegeneration, and metabolic disorders.
